Abstract

The increasing incidences of chronic inflammatory diseases such as rheumatoid arthritis and osteoarthritis have further increased the necessities for effective drug delivery systems which can enhance therapeutic value along with minimization of systemic side effects. Ability to bypass first-pass metabolism, sustained drug release and improving patient adherence has positioned transdermal system as a potent alternative for conventional oral and parenteral administration of drugs. Despite the high adherence potentials, it is known that the stratum corneum functions as a strong barrier, which severely limits transdermal delivery of many drugs. Niosome-based nanocarriers have attracted significant interest due to their biocompatibility, physicochemical stability, high drug encapsulation efficiency and the capability of enhance skin permeation and regulated drug release. This review attempts to provide an overview of the recent progress in dual-drug niosomal transdermal patches specifically planned for anti-inflammatory therapy with particular emphasis on co-delivery approach for synergistic therapeutic outcomes associated with etodolac and curcumin combination. In this review, detailed information is provided on the structural nature of niosomes, formulation elements, preparation methods, mechanism of skin permeation enhancement and also approaches for chemical and physical strategies such as microneedles (MNs), iontophoresis (IP), sonophoresis and electroporation.

Alternative drug administration techniques, such as topical (also known as dermal) and transdermal delivery systems of active pharmaceutical ingredients (APIs), are becoming increasingly significant in addition to conventional and parenteral oral dose forms. Topical and dermal formulations, on the other hand, are intended to produce systemic effects by penetrating the skin layers and achieving local effects on the skin's surface and superficial layers (such as the epidermis—topical delivery) as well as in the underlying tissues. Because other organs (such as joints, the central nervous system, hormone therapy, pain relief, etc.) might potentially be therapeutic targets, the skin/transdermal pathway is crucial for dermatological therapy[1]. Niosomes are tiny particles. Niosomes are used in targeted medication administration and have been shown in recent studies to greatly enhance transdermal drug delivery.
